I'm not sure what you mean by isolate speakers, but if the quality is not good, you may try to apply some audio processes to improve it. In Scribe > right click on the file you need to fix > Special Audio Processes > try any of the effects you have there (it depends on the problem you have in that particular file). I was unable to reproduce the issue you are reporting. Stamp always shows up the folders in alphabetical order, it does not matter if they are sorted in a different way in the Windows File Explorer. What is the size of that file? If you are getting that message, 16GB may not be enough to be able to load and edit the file. Note that a 17 hour file will consume a lot of your system resources, so you will need a powerful computer to be able to process it. In this case, I recommend you free up space on your hard drive to see if it helps. Also, if the file is stored locally on your computer, you can move it to an external hard drive and try to load it from there
